Eight people who don’t know each other are invited to Indian Island. They all think they’ve been hired for different reasons, but when they arrive on the island, they realize that none of them were actually hired and instead have been brought together by a mysterious host.

When the guests arrive on the island, they meet Mr. and Mrs. Rogers, who tell them that their host will not be there until tomorrow. That night at dinner everyone is talking about how strange it is that they don’t know anything about this mysterious “Mr. Owen.” They all agree that none of them know him or what he looks like so he must have picked them for a reason.

The guests discuss what to do. Tony Marston drinks poisoned whiskey and dies, which scares the others. They retreat to bed, where they remember their crimes and worry about dying. Vera Claythorne notices that Mr. Marston’s death is similar to a nursery rhyme in each bedroom called “Ten Little Indians.” The next morning, Mrs Rogers appears to have died in her sleep. However, Blore, Lombard and Armstrong decide that all of these deaths are murders because they can’t find anyone named Mr Owen on the island. General Macarthur feels like he’s going to die soon but goes out onto the porch before lunch anyway. Dr Armstrong finds him dead from a blow to the head later on during lunchtime

The guests are now thoroughly freaked out. They realize that one of them must be the murderer. The judge reminds everyone that they should look at all the evidence before jumping to conclusions. After breakfast, Mr Rogers is found dead with an axe in his head while chopping wood for a fire he was going to start for lunch. The remaining guests figure things out and realize that it’s probably someone from their group who is behind this madness as per the nursery rhyme requirements:

After breakfast, Emily Brent feels slightly giddy and remains alone at the table for a while. She is soon found dead with poison in her neck. Wargrave initiates an organized search of everyone’s belongings and anything that could be used as a weapon is locked away. The remaining guests sit together, passing time and casting suspicious looks at each other. Finally, Vera goes to take a bath but she is startled by seaweed hanging from her ceiling which causes her to cry out. Blore, Lombard, and Armstrong run to help her only to return downstairs to find Wargrave draped in courtroom robes with his head shot off.

That night, Blore hears footsteps in the hall. He checks to see if Armstrong is in his room, but he’s not there. They search for him and find that another Indian statue has been taken from the table.

Vera, Lombard and Blore decide to stay outside after seeing the strange things going on in the house. Blore decides to go back inside for food. He is killed when a statue falls out of a window while he is trying to enter it from the second floor. Vera and Lombard retreat to the shoreline where they find Armstrong’s drowned body on the beach. They realize that Lombard must have been responsible for Armstrong’s death because he was seen coming out of his room just before everyone went up to bed last night, so they run away together into the woods nearby. While running across an open field, they see a noose hanging from a tree branch ahead of them and feel compelled by some unknown force (the rhyme)to follow through with its instructions: “He takes her hand and she takes him along.” The two join hands as instructed by this mysterious force (the rhyme), but as soon as their hands touch, Vera shoots Lombard dead with his own gun! She returns alone to her bedroom where she finds another noose waiting for her in place of what used be there previously.
